Adrafinil primarily works as a prodrug to Modafinil, meaning that when ingested it is converted to Modafinil. Tests on elderly humans found that blood levels of Adrafinil peaked within an hour, however the neurological effects only began an hour after blood levels peaked. Based on this, researchers concluded that the nootropic benefits of Adrafinil are caused by its conversion into Modafinil.

While Adrafinil provides many of the cognitive enhancing effects commonly associated with amphetamine-like stimulants, research has shown that unlike these stimulants which produce a broad activation of neurons Adrafinil selectively activated neurons in the hypothalamus-based wakefulness circuits. Adrafinil is best taken in the morning and on an empty stomach. It is most effective at a dosage of 300 mg, although some people report needing to take 600 mg doses to experience its full benefits.
